【Abstract】Objective: To study the effects of seed soaking with different concentrations of two growth regulators, Good Fresh Harvest and Flower DuoLe, on the germination and growth of Eustoma seeds. METHODS: Using double-layer filter paper culture method and indoor potting method, the soaked seeds were cultivated in a dark incubator at a constant temperature of 25℃, and the germination rate, germination potential, germination index and growth indicators were determined. RESULTS: The two growth regulators were able to better promote the germination of Eustoma seeds.
【Keywords】 Constant temperature incubator; Platycodon grandiflorus; growth regulator; foliar fertilizer In recent years, there are reports of treatment studies to promote the germination and growth of Platycodon grandiflorus seeds from the aspects of warm water immersion, laminar treatment and chemical reagent treatment. Good Fresh Harvest is a plant growth promoter formulated with plant-based alginate as the main ingredient, supplemented with gibberellic acid and organic nutrients, and has been registered for use in leafy greens such as celery and choy sum; Long Youhua's research has shown that Good Fresh Harvest can promote seed germination by soaking maize seeds, enhance the strength of maize plants, and increase maize yield and quality. Flower DuoLuo is a concentrated growth regulator, the main components of which are boron, copper, molybdenum, zinc and other growth promoters, which can significantly improve the natural pollination ability of the crop, enhance the flowering rate, modulate the growth balance and help the crop to resist a variety of adverse natural environments and stresses.
1 Materials and methods 1.1 Test materials The seeds of Eustoma for test were provided by the Department of Chinese Herbal Medicine of Guizhou University. Test agents: Good Fresh Harvest (15% alginate water-soluble fertilizer, produced by Dongguan Spurwang Biotechnology Co., Ltd.); Flower DuoLe Plant Foliar Fertilizer: boron (B) ≥ 3.0% (chelated), copper (Cu) ≥ 0.1% (chelated), molybdenum (Mo) ≥ 0.02% (chelated), zinc (Zn) ≥ 4.0% (chelated), and other growth-promoting factors, produced by the American Shidole Group of Companies.
1.2 Experimental methods Experimental design: The experiment was set up with five concentration treatments of Good Fresh Harvest 0.025, 0.05, 0.10, 0.20, 0.40g/L; five concentration treatments of Flower DuoLe 0.05, 0.10, 0.20, 0.30, 0.40g/L; and the treatment with distilled water was used as the control (CK). The seeds of Platycodon grandiflorum with full grains were first sterilized with 0.5% KMnO4 for 30 min, then rinsed with distilled water for several times, and then soaked with 50 mL of different concentrations of Haoxianshou and Huaduole.
1.3 Data processing The data were analyzed using Kingsoft WPS Office 2012 and SPSS 12.0 statistical software.
2 Results and analysis Different concentrations of Hao Fresh Harvest seed soaking treatments of Eustoma seeds could promote the seed germination process to different degrees. Compared with the fresh water control, all treatments significantly increased the germination rate, germination potential and germination index of Eustoma seeds, and the difference with the control reached a significant level. Among them, the better effect was 0.20g/L Good Fresh Harvest, with germination rate, germination potential and germination index reaching 92%, 61% and 4.18, respectively; germination rate, germination potential and germination declined when the concentration was higher than 0.40 g/L. With the increase of the concentration of Flower DuoLe treatment, the germination rate, germination potential and germination index of Eustoma seeds showed a low-high-low trend, and the germination rate, germination potential and germination index of Eustoma seeds of all treatments were higher than that of the control, and the differences all reached a significant level.
3 Conclusion Currently, the use of plant growth regulators to regulate plant growth as an agricultural technology applied in production shows great economic potential. In recent years, while its medicinal and health care values have been widely accepted, wild resources have long been unable to meet the demand, and researchers have carried out studies on the artificial cultivation of Platycodon grandiflorum from seed propagation or asexual cuttings propagation, sowing amount, sowing date, harvesting period, and centering and rectification, and so on. And this experimental study shows that the use of growth regulators can better promote the germination of Eustoma seeds and regulate the growth of Eustoma plants, improve the economic benefits of Eustoma, and provide a certain technical basis for the high-yield, high-efficiency, high-quality cultivation mode of Eustoma, which is of great significance as a guide to production.
